The COVID-19 has now been declared a global pandemic by the World Health Organization. No approved drug is currently available therefore an urgent need for the development any potential antiviral therapeutics for prevention and treatment of COVID-19. Many scientific research to be spotlights and efforts are being made to provide potential therapeutics. The major objectives of the present research is to apply this incredible in-silico screening methodology for the official Siddha formulation Kabasura Kudineer against SARS- CoV-2 3CLpro.

Kabasura Kudineer (KK), an official Siddha formulation comprises of 15 medicinal plants namely Adhatoda vasica, Andrographis paniculata, Anacyclus pyrethrum, Coleus ambonicus, Cypreus rotundus, Clerodendrum serratum, Hyfrophila auriculata, Saussurea lappa, Sida acuta, Syzygium aromaticum, Terminaila chebula, Tinospora cordifolia, Tragia involuerta, Zingiber officinale, and Piper longum has proven clinical track record in management of viral infections.

The present study selected 145 active compounds from Kabasura Kudineer  based on the biological activities. Identified 15 highly potential  therapeutic compounds : Acetoside, Luteolin 7 -rutinoside, rutin, Chebulagic acid, Syrigaresinol, Acanthoside, Violanthin, Andrographidine C, myricetin, Gingerenone -A, Tinosporinone, Geraniol, Nootkatone, Asarianin and sitosterol are main compounds from Kabasura Kudineer  which may inhibit 3CLPro. Several active sites were identified in Coronavirus. Highest binding energy were seen in Kabasura kudineer compare to Synthetic drugs (Azithromycin,Hydroxychloroquine, Ivermectin,Lipinavir, Oseltamivir and Ritonavir)  showed minimal binding capacity. Kabasura kudineer may be exert against SARS-CoV-2 by either blocking the host cell receptor /inactivating 3CLpro.  Potential compounds from Kabasura kudineer showed effective antiviral therapeutics for the prevention of COVID-19 and other newly emerging viral infection  in future.

Major recommendation of the findings; Emerging Mutant virus is unstoppable including MERS-COV;  SARS- CoV and SARS CoV-2 and Fast-spreading UK-type variants of SARS-CoV-2 etc.. It’s a high time we should integrate all the treatment system in our country including Ayurveda, Yoga and Naturopathy, Unani, Siddha and Homeopathy for the control of emerging viruses. We suggested that Kabasura kudineer –has  potential compounds may be used to develop effective antiviral therapeutics for the prevention of COVID-19 and also newly emerging viruses. We have strengthen health system in our country use 50% traditional herbal medicine and 50 % allopathy. Kabasura kudineer should be appropriately evaluated and  concern to maintain the original quality of the products.
